Azospirillum brasilense is a plant growth promoting rhizobacterium which colonizes a large number of agronomically important grasses and promotes their growth through phytohormone production and nitrogen fixation. The genome of A. brasilense encodes 23 sigma factors including 10 extra-cytoplasmic function (RpoE) sigma factors and 5 RpoH sigma factors. In order to understand the role of each of these sigma factors in its physiology, we are knocking out each gene encoding alternative sigma factor and trying to characterize their phenotype and identify members of their regulon using proteomic and transcriptomic approaches. We have shown that oxidative and photo-oxidative stress in A. brasilense is regulated by a network of multiple cascades of RpoE and RpoH sigma factors. We are also investigating the role of genes involved in the transport of dicarboxylates, glucose and ethanol, and their regulation by alternative sigma factors. We have determined whole genome sequence of the four Indian isolates of A. brasilense to identify their unique features vis a vis their South American cousins. Currently, we are also focusing our attention on the role of six Iron starvation sigma factors (FecI) in siderophore mediated Iron nutrition. We are using our understanding of the systems biology of A. brasilense to produce high value phytochemicals (amorphadiene, geraniol etc) in A. brasilense using synthetic biology approach.
